Abstract

Chelating azides accelerate and enhance the efficiency of copper-catalyzed azide–alkyne cycloadditions (CuAAC), showing promise for intracellular applications. Spectroscopic studies showed that abundant thiol-containing glutathione (GSH) hinders the CuAAC reaction of chelating azides at stoichiometric GSH-to-CuI ratios by sequestering Cu in the form of CuAAC-inactive CuI-thiolate clusters.
