New paradigm for potency : The crystal structures of E.coli IspH in complex with (E)-4-mercapto-3-methylbut-2-en-1-yl diphosphate enable us to design IspH ligands with improved binding capacities. A novel binding feature gave rise to compounds that present a new paradigm for the development of more potent IspH inhibitors.
